Reagents and materials:โ
- Ethanol 88% 600-700 mlโ
- Powder black pepper 300 gโ
- 24 g Potassium hydroxide KOHโ
- 1600 ml distilled waterโ
- Paper filters diameter 180 mmโ
- Flask 2000 mlโ
- Beakersโ
- Funnelโ
- Heaterโ
Place 300 g of black pepper powder and 500 ml of 88% EtOH into a 2000 ml flask. Fig 1โ
Fig 1โ
The mixture is boiled under a reflux condenser for 5-7 hours. Fig 2โ
Fig 2โ
A solution of potassium hydroxide is prepared: 24 g Potassium hydroxide KOH + 200 ml water. Fig 3โ
Fig 3โ
The piperine extract in ethanol is filtered warm (40-50โ) into the prepared potassium hydroxide solution (if there is a deficiency of ethanol, more than half of the solvent can be distilled off and saved before mixing with the potassium hydroxide solution). The addition of potassium hydroxide or another alkali is necessary to remove resins. Fig 4โ
Fig 4โ
Powder of pepper is additionally washed with 100-200 ml of warm (40โ) ethanol on a filter. Fig 5โ
Fig 5โ
The resulting filtered solution is heated to 60-70โ. Fig 6โ
Fig 6โ
The heated solution is transferred to a large 2000 ml beaker. Fig 7โ
Fig 7โ
Water is added to the solution until sediment formation stopped (~1400 ml water). Fig 8โ
Fig 8โ
The resulting suspension is cooled. Fig 9โ
Fig 9โ
The piperine precipitate is filtered on filters. Piperine is much dispersed and passes through the pores of most filter materials. Fig 10โ
Fig 10โ
The piperine precipitate is washed with water until a neutral reaction is achieved (it is necessary to remove the alkali). Fig 11โ

Piperine is dried in a desiccator without access to air and sunlight. Fig 12โ
